چکیده
Microphthalmia-associated transcription factor (MITF) plays a pivotal role in melanocyte development and function as well melanoma pathogenesis. MITF amplification is common has been associated with resistance to targeted therapies. Melanin, whose synthesis largely regulated by MITF, powerful anti-oxidant activity. Here we show that additionally regulates transcriptional antioxidant program increases survival of cell lines protecting the cells from reactive oxygen species (ROS)-induced damage. In addition, this redox correlates expression human patient-derived samples. A zebrafish model verified decreases ROS-mediated DNA damage vivo. Some target genes, such IDH1 NNT, are through direct binding canonical E-BOX sequences proximal their promoters. its genes reduce cytosolic mitochondrial ROS cells. Collectively, these data suggest major driver cellular state may offer potential novel therapeutic approaches melanoma.

miR-148 Regulates Mitf in Melanoma Cells
The Microphthalmia associated transcription factor (Mitf) is an important regulator in melanocyte development and has been shown to be involved in melanoma progression. The current model for the role of Mitf in melanoma assumes that the total activity of the protein is tightly regulated in order to secure cell proliferation. Previous research has shown that regulation of Mitf is complex and inv...

Microphthalmia-associated transcription factor (MITF), a key regulator of melanocyte development, belongs to the class of basic-helix ⁄ loop ⁄ helixleucine-zipper transcription factors. Microphthalmia-associated transcription factor regulates multiple genes related to melanin synthesis (e.g. TYR, DCT), control of apoptosis (e.g. BCL2) and cell cycle progression (e.g. p21, p16, CDK2, TBX2) as we...
متن کاملA Conditional Zebrafish MITF Mutation Reveals MITF Levels Are Critical for Melanoma Promotion vs. Regression In Vivo
The microphthalmia-associated transcription factor (MITF) is the "master melanocyte transcription factor" with a complex role in melanoma. MITF protein levels vary between and within clinical specimens, and amplifications and gain- and loss-of-function mutations have been identified in melanoma. How MITF functions in melanoma development and the effects of targeting MITF in vivo are unknown bec...

Melanoma is one of the most dangerous and lethal skin cancers, with a considerable metastatic potential and drug resistance. It involves a malignant transformation of melanocytes. The exact course of events in which melanocytes become melanoma cells remains unclear. Nevertheless, this process is said to be dependent on the occurrence of cells with the phenotype of progenitor cells - cells chara...
متن کاملPRMT5 Is Upregulated in Malignant and Metastatic Melanoma and Regulates Expression of MITF and p27Kip1
Protein arginine methyltransferase-5 (PRMT5) is a Type II arginine methyltransferase that regulates various cellular functions. We hypothesized that PRMT5 plays a role in regulating the growth of human melanoma cells. Immunohistochemical analysis indicated significant upregulation of PRMT5 in human melanocytic nevi, malignant melanomas and metastatic melanomas as compared to normal epidermis. F...
